Isolate® Sperm Separation

Description / Use

Isolate® is a superior medium for separation of the motile fraction of sperm from its surrounding seminal plasma by density centrifugation.

As a two-layer gradient system, ISolate effectively removes cellular contaminants-dead sperm, white cells, and miscellaneous debris-yielding a sample that contains predominately motile sperm.

ISolate is a sterile, colloidal suspension of silica particles stabilized with covalently bound hydrophilic silane is a HEPES-buffered HTF. This buffered system maintains a physiological pH at ambient conditions (do not use this product in a CO2 incubator unless tightly capped).

a fresh or cryopreserved semen sample is overlaid on 1.5 to 2.0mL aliquots of the two-layer gradient system in a conical centrifuge tube. The prep tube is then centrifuged at a low speed, ~300xg for 20 minutes. This action causes seperation of the seminal components. Dead sperm and debris are found in the upper portion of the tube while a fraction of highly motile, morphologically normal cells will form a pellet in the bottom.

Irvine Scientific has conducted extensive performance and safety testing on ISolate. These studies indicate that it is comparable or superior to the standard separation techniques utilizing Percoll® and superior in performance to standard swing-up procedures.

Sperm Cryopreservation Media

Description / Use

Media containing Test-yolk buffer (TYB) has been shown to be effective in preserving sperm when refrigerated for short intervals of time, and for long term frozen storage with the addition of glycerol.
